One of those mechanisms includes the thymine DNA-glycosylase (TDG) and the downstream factors of DNA base excision repair (BER), which were shown to be involved in the dynamic, locus-specific regulation of DNA methylation. The underlying mechanisms to ensure this necessary epigenetic plasticity are therefore active and highly dynamic in embryonic stem cells (ESC). Once you get the technique down, you can skip to building both right and left limbs at the same time as each limb’s parts are in close proximity on the sprue tree. For example: the instructions tell you how to build one limb at a time, a five step process of foot, lower leg, knee, upper leg and thigh (or hand, forearm, elbow, upper arm, and shoulder). The process is almost identical on all three – challenging at first, but easier as you go. By that I mean assembling the arms and legs, the most time consuming build on each kit.
